Hackaday: Learn 15 Print-in-Place Mechanisms In 15 Minutes. “3D printed in-place mechanisms and flexures, such as living hinges, are really neat when you can get them to print correctly. But how do you actually do that? YouTuber [Slant 3D] is here with a helpful video demonstrating the different kinds of springs and hinges (Video, embedded below) that can be printed reliably, and discusses […]

“…organisms are nothing like mechanisms, and…they have fascinating properties that cannot be explained by a complicated machine, however complicated. They can only be explained by complex systems. The distinction is this. A Harrier jet engine is a complicated machine, but a small organism, a single-cell amoeba, is a complex system. And the difference is either they have re-entrant loops that can't be mapped, they are never fully determined or determinable, not entirely predictable. But that doesn't mean there's chaos. There is a sweet spot in everything between order and disorder. And it is that at which all existence seems to settle, and particularly life.”

Prototype for a new turntable switch array is promising. Copper contacts on flexible plastic make a circuit with a fixed bar of copper.
I still need to wire up the rest of the switches, but my soldering iron broke somewhere along the line so need to wait for a replacement to arrive.
I'm hoping these home made switches are effective and robust enough to make into a final version. Needs to be a bit smaller overall though.

Draft continental #strategy on free movement developed

Free Movement of Persons is an integral part of the #African #integration and along with the African #Passport is one of the flagship projects of #Agenda #2063.

Pursuant to this goal of integration, in January 2018 the Protocol to the Treaty establishing the African #Economic #Community relating to free movement of persons, right of residence and right of establishment was adopted by the #AU Assembly. This was followed in March 2018 with the Agreement Establishing the African Free #Trade Area, the Protocol on Trade in #Goods, the Protocol on #Trade in #Services and the Protocol on Rules and #Procedures on the #Settlement of #Disputes.

As a follow up to this, the #AfricanUnion (AU) and the Regional Economic Communities ( #RECs ) have developed a draft joint continental strategy to accelerate the Right of Entry for the Protocol to the treaty establishing the African Economic Communities relating to the Free Movement of Persons, Right of Residence and Right of Establishment. The strategy aims to ensure that RECs and Member States popularise and promote the adopted African Passport as one of the key #mechanisms through which free movement of persons will be realised.

Excellent talk by Iain McGilchrist, with so many deep insights into how "mechanisms" destroy personal authenticity, common sense, spontaneity and humanity in our lives. Deeply true.

We must fight to maintain the remnants of unmechanised interaction. In our work, in our personal lives.

Mastodon to me is one small but important part of that battle.

NatureMolecular mechanisms of SARS-CoV-2 resistance to nirmatrelvir - NatureNirmatrelvir is a specific antiviral targeting the main protease (Mpro) of SARS-CoV-2, and has been approved to treat COVID-191,2. As an RNA virus characterized by high mutation rates, whether SARS-CoV-2 will develop resistance to nirmatrelvir is a concern. Our previous studies have shown that several mutational pathways confer resistance to nirmatrelvir but some result in a loss of viral replicative fitness, which in turn are compensated by additional mutations3. The molecular mechanisms for this observed resistance are yet unknown. Here we combined biochemical and structural methods to demonstrate that mutations at the substrate binding pocket of the Mpro can allow SARS-CoV-2 to develop resistance to nirmatrelvir in two distinct ways. Comprehensive studies of 14 complex structures of Mpro mutants with drugs or substrate revealed that mutations at the S1 and S4 subsites significantly decreased inhibitor binding, while mutations at the S2 and S4’ subsites unexpectedly increased protease activity. Both mechanisms contributed to nirmatrelvir resistance, whereas the latter compensated for the loss in enzymatic activity of the former, which in turn accounted for the restoration of viral replicative fitness as we have observed previously3. Such a profile was also observed for ensitrelvir, another clinically relevant Mpro inhibitor. These results shed light on the mechanisms by which SARS-CoV-2 evolves to develop resistance to the current generation of protease inhibitors and provide the basis for the design of next-generation Mpro inhibitors.
